The phylogenetic relationships of Mytilarioideae Chang and allied subfamilies of Hamamelidaceae were studied based on sequences of the ITS regions of nrDNA and the matK and rbcL genes of cpDNA. The ITS and matK sequences were determined from 8 taxa to reconstruct phylogenies of Mytilarioideae and Hamamelidaceae. A combined analysis of the ITS and matK data was conducted to better estimate an integrated molecular phylogeny of the subfamily and the family. Phylogenetic trees generated from the ITS and matK, as well as rbcL sequences (from GenBank) were compared with each other. All molecular phylogenies obtained in this study based on different data sets supported that the Mytilarioideae is a monophyletic group. The subfamily Liquidambaroideae is basal within the family, and 4 genera from other different subfamilies, i.e., Hamamelis and Disanthus, and Exbucklandia and Rhodoleia, formed two pairs of sistergroup, respectively.
