XVI International Botanical Congess
This study was intended to demonstrate the usefulness of nuclear ribosomal DNA ITS sequences for resolving phylogenetic relationships within the several genera(Apiaceae). The internal transcribed spacer region(ITS1 and ITS2) of the 18S-25S nuclear ribosomal DNA sequences and the intervening 5.8S region were sequenced from 13 taxa of Apiaceae subfamily Apioideae five genus(Angelica, Libanotis, Lingusticum, Peucedanum, Pimpinella). We found length variation the ITS1 varied from 203 to 218bp, the ITS2 varied from 211 to 224bp, and the 5.8S was 163bp. Divergence values in pairwise comparison among all taxa were ranged from 0 to 0.213 of nucleotides in ITS1 and from 0 to 0.225 in ITS2. A highly resolved strict-consensus tree was obtained that showed the phylogenetically useful information of ITS sequences from several genera.
