PRODUCTION OF NATURAL PRODUCTS FROM HAIRY ROOT CULTURES OF MEDICINAL PLANTS

We successfully established hairy roots of seven medicinal plants by cocultivation on sterile plantlets with Agrobacterium rhizogenes A4, ATCC15834. Transformed roots were confirmed that hairy roots examined contain both TL-DNA and TR-DNA region of Ri-plasmid by PCR amplification analysis of DNA. Hairy root lines grew rapidly with extensive lateral branching in growth regulator-free medium. To determine the optimal medium for growth of hairy roots, the effect of eleven basal media were investigated. The production of secondary metabolites in hairy root cultures were influence by the addition of chitosan, biotic and abiotic elicitors to the medium. For commercial production of natural products, hairy roots were cultured in 15L air-lift type bioreactor.
